Immune Cells Get Cancer-Fighting Boost from Nanomaterials
Researchers used bundled carbon nanotubes (CNTs) to incubate cytotoxic T cells, a type of white blood cell that is important to immune system functions. The topography of the CNTs enhances interactions between cells and long-term cultures, providing a fast and effective stimulation of the cytotoxic T cells that are important for eradicating cancer. Cerebral Ischemia Increases Bone Marrow CD4+CD25+FoxP3+ Regulatory T Cells in Mice via Signals from Sympathetic Nervous System
Recent evidence has shown that an increase in CD4+CD25+FoxP3+ regulatory T cells may contribute to stroke-induced immunosuppression. Scientists used a transient middle cerebral artery occlusion model in mice and specific pathway inhibitors to demonstrate that stroke activates the sympathetic nervous system, which was abolished by 6-OHDA. Role of Tyk-2 in Th9 and Th17 Cells in Allergic Asthma
In a murine model of allergic asthma, researchers found that Tyk-2(−/−) asthmatic mice have induced peribronchial collagen deposition, mucosal type mast cells in the lung, interferon-regulatory factor 4 and hyperproliferative lung Th2 CD4+ effector T cells over-expressing IL-3, IL-4, IL-5, IL-10 and IL-13. Brain Antigen Targeted in Paraneoplastic Neurologic Disease Limits Neuronal Autoimmunity but Not Cancer Rejection by Adoptively Transferred T Cells
In a C57BL/6 mouse model, central nervous system (CNS) antigen expression limited antigen-specific CD4+ and CD8+ T-cell expansion. CNS antigen expression did not limit tumor rejection by adoptively transferred transgenic T cells but did limit the generation of a memory population that could be expanded upon secondary challenge in vivo. DRB1*0402 May Influence Arthritis by Promoting Naive CD4+ T-Cell Differentiation in to Regulatory T Cells
HLA-DRB1*0401 expression in humans has been associated with a predisposition to developing rheumatoid arthritis and collagen-induced arthritis, while HLA-DRB1*0402 is not associated with susceptibility. Investigators determined if mice transgenic for human *0401 have a CD4+ T-cell repertoire that predetermines pro-inflammatory cytokine production. [Eur J Immunol] Abstract

Systemic CD8+ T Cell-Mediated Tumoricidal Effects by Intratumoral Treatment of Oncolytic Herpes Simplex Virus with the Agonistic Monoclonal Antibody for Murine Glucocorticoid-Induced Tumor Necrosis Factor Receptor
Investigators examined the tumoricidal effects of oncolytic HF10, a naturally occurring mutant of herpes simplex virus type-1, combined with an agonistic DTA-1 monoclonal antibody specific for the glucocorticoid-induced tumor necrosis factor receptor. An accumulation of CD8+ T cells, including tumor- and herpes simplex virus type-1-specific populations, and a decrease in the number of CD4+ Foxp3+ T regulatory cells were seen in both HF10- and DTA-1-treated tumors. CD4+CD25+ Regulatory T Cells Are Not Required for Mesenchymal Stem Cell Function in Fully MHC-Mismatched Mouse Cardiac Transplantation
By using C57BL/6 mouse mesenchymal stem cells (MSCs) as donor-derived or recipient-derived or as third-party MSCs, scientists discovered that MSCs suppressed CD4+ T cell proliferation and prolonged mouse cardiac allograft survival in a dose-dependent and non-major histocompatibility complex (MHC)-restricted manner. Ursolic Acid Ameliorates Autoimmune Arthritis via Suppression of Th17 and B Cell Differentiation
Scientists examined the effects of ursolic acid (UA) on collagen-induced arthritis (CIA) in mice, and identified the mechanisms underlying the effects. UA treatment significantly reduced the incidence and severity of CIA-induced arthritis, accompanied by decreased expression of proinflammatory cytokines and oxidative stress markers in arthritic joints. The Injury-Induced Myokine Insulin-Like 6 Is Protective in Experimental Autoimmune Myositis
Researchers analyzed the role of insulin-like 6 (Insl6) in experimental autoimmune myositis. Insl6 deficiency in mice led to a worsened myositis phenotype including increased infiltration of CD4 and CD8 T cells and the elevated expression of inflammatory cytokines.
