FoxP3+ROR(Gamma)t+ T Helper Intermediates Display Suppressive Function against Autoimmune Diabetes
In this study, researchers report that FoxP3+ROR(gamma)t+ intermediates arise in the NOD mouse T cell repertoire prior to inflammation and can be expanded with tolerogen without further differentiation. [J Immunol]

Dermatology: Watching Immune Cell Movement To and From the Skin
A team of researchers has now used mice engineered to express the photoconvertible fluorescence protein Kaede, which changes from green to red when exposed to violet light, to track regulatory T cell movement under physiologic conditions and during immune responses in the skin. [Press release from ScienceDaily discussing online prepublication in J Clin Invest]

In Vivo Requirement for Atg5 in Antigen Presentation by Dendritic Cells
Mice with dendritic cell-conditional deletion in Atg5, a key autophagy gene, showed impaired CD4+ T cell priming after herpes simplex virus infection and succumbed to rapid disease. [Immunity]

Activated Regulatory T Cells are the Major T Cell Type Emigrating from the Skin During a Cutaneous Immune Response In Mice
Results suggest that regulatory T cells in circulation infiltrate the periphery, traffic to draining lymph nodes, and then recirculate back to the skin, contributing to the downregulation of cutaneous immune responses. [J Clin Invest]

Competing Feedback Loops Shape IL-2 Signaling Between Helper and Regulatory T Lymphocytes in Cellular Microenvironments
Researchers combined mathematical modeling and experiments to dissect the dynamics of the interleukin-2 (IL-2) signaling network that links the prototypical IL-2 producers, conventional T helper cells, and regulatory T cells. [Proc Natl Acad Sci USA]

Expression of Helios, an Ikaros Transcription Factor Family Member, Differentiates Thymic-Derived from Peripherally Induced Foxp3+ T Regulatory Cells
Helios, a member of the Ikaros transcription factor family, is preferentially expressed at the mRNA level by regulatory T cells. Researchers evaluated Helios protein expression using a newly generated monoclonal antibody and demonstrated that it is expressed in all thymocytes at the double negative 2 stage of thymic development. [J Immunol]

A Regulatory Role for Macrophage Class A Scavenger Receptors in TLR4-Mediated LPS Responses
Researchers have revealed a mechanism where scavenger receptors regulate the activation status of the immune system and are involved in balancing a proper immune response to infection. This regulation could also be important in maintaining tolerance since these receptors have been shown to be involved in regulation of self reactivity. [Eur J Immunol]

Foxp3-Transduced Polyclonal Regulatory T Cells Suppress Natural Killer Cell Functions in a TGF-Beta Dependent Manner
The purpose of this study was to investigate the suppressive effect of Foxp3-transduced CD4+ T cells on natural killer (NK) cell functions, and thus to present a possible method for managing NK cell-related autoimmune abnormalities. [Autoimmunity]

IL-10 is Required for Human CD4+CD25+ Regulatory T Cell-Mediated Suppression of Xenogeneic Proliferation
In this study, researchers analyzed whether interleukin-10 (IL-10) is required for regulatory T cells to suppress xenogeneic responses in vitro by small interfering RNA-mediated IL-10 knockdown. [Immunol Cell Biol]

Elevated Expression of Foxp3 in Tumor-Infiltrating Regulatory T Cells Suppresses T Cell Proliferation and Contributes to Gastric Cancer Progression in a COX-2-Dependent Manner

Data demonstrate a mechanism by which tumor-infiltrating regulatory T cells with increased Foxp3 expression can mediate immune suppression via COX-2/PGE2 production in the gastric cancer microenvironment. [Clin Immunol]

Acorda Therapeutics 4Q Loss Widens, as Costs Grow
The Hawthorne, N.Y., company said it lost $22.5 million, or 59 cents per share, in the three months that ended Dec. 31. That compares with a loss of $20.2 million, or 54 cents per share, a year ago. [BusinessWeek]

Idera Pharmaceuticals Presents Mechanism of Action Data on IMO-3100, a Novel Toll-like Receptor Antagonist for Autoimmune Diseases, at Keystone Symposium
Idera Pharmaceuticals, Inc. presented data on the mechanism of action of IMO-3100, an antagonist of Toll-like Receptor (TLR) 7 and TLR9, in a preclinical primate model at the Keystone Symposia conference “Tolerance and Autoimmunity”. [The Wall Street Journal]

CEL-SCI Study Shows CEL-2000 Vaccine Blocks Progression of Rheumatoid Arthritis
CEL-SCI Corporation and their scientific collaborators announced today that the Company’s CEL-2000 vaccine demonstrated that it is able to block the progression of rheumatoid arthritis in a mouse model. [CEL-SCI Corporation Press Release]

Five Prime Therapeutics, Inc. Announces Milestone for Therapeutic Program
Five Prime Therapeutics, Inc. announced that it received a milestone payment from Centocor Research & Development, Inc. for the selection of a therapeutic that will be pursued for immunology-related indications. [Five Prime Therapeutics Press Release]

New Study Demonstrates Effectiveness of Prochymal as a Rescue Therapy for Pediatric Patients with Severe Graft vs. Host Disease
Osiris Therapeutics, Inc. reported that a new study shows Prochymal achieved an overall response rate of 63% when used as a rescue therapy in children suffering from severe treatment resistant graft vs. host disease. [Osiris Therapeutics Press Release]
